The collision took place on the M6 Junction 12 North for Gailey, with paramedics called to the scene at around 5.57am.
A man, the driver of the car, has been taken to hospital with potentially serious injuries.
Another man, who was the driver of the lorry, was treated by ambulance staff and discharged at the scene.
As a result, traffic was stopped between Junction 11a for the M6Toll road and Junction 12 for the A5 for Telford and Staffordshire.
It caused one hour delays on the M6 northbound as well as three miles of congestion.
A spokeswoman for West Midlands Ambulance Service, said: "We were called at 5.57am to a road traffic collision involving a car and a lorry on the M6 Junction 12 North, Gailey in Stafford.
"Two ambulances and two paramedic officers attended the scene. Upon arrival, we found two men who were the drivers of the vehicles.
"The first man was the driver of the car. He was assessed and had potentially serious injuries. He received treatment from ambulance staff at the scene before being conveyed to New Cross Hospital for further assessment.
"The second man was the driver of the lorry. He was assessed and had sustained minor injuries. He received treatment from ambulance staff followed by self-care advice and was discharged at the scene."
As of around 9.18am, the incident had been cleared with all lanes having re-opened.
Motorists are still expected to experience around 45 minutes of delays on approach to the scene of the crash.
